The S100 family of proteins are found in vertebrates and are characterized by two calcium-binding sites that have a helix-loop-helix conformation. The family of 24 is divided into three subgroups based on function: those with intracellular, intracellular plus extracellular, and extracellular regulatory effects. The calcium-binding protein S100A8, also called MRP8 or Myeloid-related protein 8, and Calgranulin A, forms a heterodimer with S100A9, called Calprotectin. The heterodimer is expressed by granulocytes, monocytes, and early differentiation states of macrophages, and is known to be an important pro-inflammatory mediator in acute and chronic inflammation. S100A8 and 9 are DAMPs, or Damage-associated molecular pattern molecules, and are able to activate receptors of the innate immune system, one being TLR4.
