<b>Anti-CD11b staining of murine neutrophils.</b> <i>Ex vivo</i> murine (C57BL6/J) neutrophils from zymosan-elicited peritoneal exudate were stained with anti-CD11b antibody and analysed by flow-cytometry. Forward- and side-scatter was used to gate on neutrophil population (data not shown) and fluorescence intensity analysed. Left panel shows the signal obtained from cells pre-treated with Fc-blocking reagent and subsequently stained with anti-CD11b (hybridoma-derived antibody obtained from competitor- clone M1/70) directly conjugated to PerCP. Comparison to unstained cells shows that that cells analysed are predominantly CD11b-positive. Right panel shows staining first with anti-CD11b (24-188- clone 5C6 (recombinant version)) or isotype control (.1) followed by staining with fluorescently conjugated secondary antibody (goat anti-rat). As a further control- cells were also only stained with secondary antibody. Comparison of the staining pattern obtained using 24-188 compared to isotype- and secondary-only-control shows that the analysed cells are predominantly Cd11b-positive- agreeing with findings obtained using hybridoma-derived antibody.
